Chemical libraries in the SMSC:

ChemBridge (ChemBridge Corp.): 34,000 compounds selected from ~ 1,000,000 to create maximum chemical diversity. www.Hit2Lead.com/

LOPAC (Sigma-RBI): Library of Pharmacologically Active Compounds. 1280 compounds including marketed drugs, failed development candidates and “gold standards” with well-characterized activities. www.sigmaaldrich.com

Spectrum (MicroSource): Spectrum (MicroSource): 2000 compounds, ~20% overlap with LOPAC. US Drug Components (50%), International Drug Components (10%), natural products and other bioactive components (40%). www.msdiscovery.com/

The Small Molecule Screening Core owns four chemical libraries based on two different principles.
The bigger collection is the diverse historical library of 34,000 compounds manufactured by ChemBridge Corporation (San Diego, CA). The library consists of handcrafted drug-like organic molecules with molecular weight in a range of 250 – 550, which were dissolved in DMSO at concentration 5mg/ml or 10-20 mM depending on molecular weight. Their structure and >95% purity have been validated by NMR. The library is formatted in 96-well format with 80 compounds per plate (16 wells contain only DMSO and can be used for controls). In this library often only a single representative of a series of analogs is retained. The idea is to retain as much chemical diversity as possible with the fewest number of compounds.

We also have two libraries of pharmacological compounds: 1280 compounds in LOPAC1280 library from Sigma and 2000 compounds in Spectrum collection from Microsource. Compounds in these libraries are dissolved in DMSO, in 10mM concentration (MW is < 500). Most of the compounds are FDA or internationally approved drugs, experimental bioactive or pure natural products. There is <20% overlap between these two libraries. SMSC recently acquired a new library of 480 compounds exclusively composed of pure natural products as lead identifying materials (compounds are dissolved in DMSO at 5mM).
